CCAAT/enhancer binding proteins (C/EBPs) are a family of six proteins. Of the isoforms, the C/EBP-α and C/EBP-β isoforms are widely expressed and well studied. The expression of the C/EBPs is regulated at multiple levels during several physiological and pathophysiological conditions through the action of a range of factors, including hormones, mitogens, cytokines, nutrients and certain toxin. The role of C/EBP-α seems to be purely antiproliferative, and it is likely that this protein will emerge as a general tumor suppressor in both hematopoietic and nonhematopoietic cell types. By contrast, C/EBP-β might both inhibit and promote cell cycle progression, depending on the cellular context and C/EBP-β forms present. Association of C/EBPβ with NF-κB family members has been demonstrated in inflammatory responses. Sumoylation appears to be inhibitory for C/EBPα and C/EBPβ. Acetylation of C/EBPβ affects association with co-regulators such as histone deacetylase, and can present either inhibitory or activating effects. |
